JAK inhibitors long term efficacy?

After failing all the biologics available I am trying JAK inhibitors. I am well aware that those are not as safe as anti-TNFs but it is my last chance to get my disease under control. At least until new drugs appear on the market.

I would like to know if these JAK's lose efficacy over time as well. It is an information I could not find anywhere, so I would appreciate any insight on it. Thank you.
 
I don't know the answer to your question, but if they do lose efficacy over time it will be for a different reason than why the biologics commonly lose efficacy. Biologics and other large protein drugs most commonly lose efficacy because they are immunogenic and stimulate the recipients' immune system to produce antibodies that react with and block the beneficial action of the drug. JAK inhibitors, by contrast, are small molecules and thus unlikely to trigger the development of any antibodies.

While nothing is certain in biological systems, and there are always exceptions to every rule, my best guess is that JAK inhibitors are unlikely to lose effectiveness over time.

Thank you very much Scipio for your reply. It is good to know that JAKs do not produce antibodies since this is my main concern with this new treatment.
